The dual laser nature of the Arcturus XT microdissection system means ultimate flexibility for tissue prep: a wide variety of tissues can be mounted on - and subsequently microdissected off of - multiple slide types. In a previous Tip Tuesday we covered methods for choosing which slide type (plain glass, glass membrane, framed membrane) is most appropriate for various applications; today we will cover the PEN membrane framed slides in greater depth.
Framed slides can be particularly useful for researchers working with exceptionally thick, uneven, or hard tissue - undecalcified bone is a great example here. In addition, GFP-labeled tissue that can't be dehydrated due to delocalization of the GFP signal can greatly benefit from use of frame membrane slides.
Framed membrane slides offer a fast, precise, non- contact method of microdissection. Attached is a protocol for mounting tissue and using a plain glass slide underneath for support. Be sure to check it out!
